Bio 124 Evolution and Organismal Diversity Winter 2012 Week 4 Lab PTC gene and Hardy-Weinberg Lab Overview: Week 4 Lab: Gel Electrophoresis and Lesson on Determining PTC genotype Week 4 Recitation: Analysis of Results Week 6 Lab: Lab report DUE to Lab TA Procedure: Conducting Gel Electrophoresis and Lesson on Determining PTC genotype A. Gel Electrophoresis 1. Carefully unwrap your gel and place it in the gel rig. Have you TA come check your set up to make sure your gel and rig are oriented correctly. 2. Fill the reservoir chamber with 1X TBE electrophoresis buffer in sufficient volume to adequately cover the entire gel by ½ cm. 3. If there are bubbles in the wells, gently tap the bottom of the gel rig to remove the bubbles. 4. Use a micropipette with a fresh tip to load 20 µL of pBR322/BstNI size marker into the second lane from the left (lane #2). 5. For each student, add approximately 10 µL of the undigested (U) and approximately 15 µL of the digested (D) samples into the different wells (#3-10). You should add the samples from each student (U and D) next to each other.
