Learning objectives At the end of this lecture, you will be able to: 1) Use a Southern blot to identify the “C-type” RFLP fragment linked with Huntington’s disease. 2) Explain how the mutant Huntingtin protein leads to disease. 3) Define genomics and (4) describe the process of shot-gun sequencing. 5) Describe the identification of novel genes using the process of scanning for Open Reading Frames and generating expressed sequence tags.

1. A note about DNA probes : = A radioactively or chemically labeled single- stranded fragment of a known DNA sequence that binds to and thus detects its complementary sequence in a sample containing many different sequences. Thus, a DNA probe that binds to a complementary DNA sequence found here ! ! can bind to the same complementary DNA sequence found on a homologous chromosome here. This DNA probe will not bind to non- complementary DNA sequences found elsewhere, like here, or over here.
