1 BSCI 410-Liu /Chang Homework #3 (50 pts) Fall 2011 Due: Tuesday (Nov. 8) at 11:00 AM at the beginning of class Review of gene structure. 1. Given the following diagram of a genomic DNA region containing 1 gene, a. Which parts of the DNA (A thru K) will be present in the mRNA? Use the letters A-K in your answer. (3 pts) b. Which parts of the DNA (A thru K) will be present in cDNA? Use the letters A-K in your answer. (2 pts) c. Which segments contain coding sequences? Use the letters A-K in your answer (1 pt) Methods in functional genomics / forward and reverse genetics. 2. Suppose you discover a new recessive hereditary disorder in humans called wrinkled feet (wf). By genetic mapping, you find that the gene responsible for this disorder is located somewhere within a particular 200 kb genomic region. The DNA in the 200 kb genomic region has been cloned, but the WF gene itself has not been identified. a. Suppose you find that the human sequence is poorly annotated in this genomic region, so you do not know how many genes there are nor where the coding sequences lie within the 200 kb fragment. List 3 general methods discussed in lecture that you could use to identify the gene- encoding sequences that lie within this cloned 200 kb genomic region. (3 pts) b. Suppose the defect is due to a 2 kb deletion in the WF promoter sequence. Briefly explain how you could identify the WF gene using each of the three methods in your answer to part (a). In your explanation, please include a description of what the data would look like and how it
