January 26, 2010 Paul Gonzales Lab Session: 49540 Bio206L 2.1 Objective Magnification Total Image Magnification (with 10X ocular) Numerical Aperture (NA) Resolution if λ =500 nm 4X objective 40X .1 3060 μ m 10X objective 100X .25 1224 μ m 40X objective 400X .65 471 μ m 100X objective 1000X 1.25 245 μ m All numbers match with the exception of the numerical aperture for the 100X objective. While online says a value of 1.4 is the NA, it was actually 1.25 on our microscopes. 2.2 (10X objective 10X = 100X mag.) Paramecium 2.3 - Ensure 100X total magnification and condenser raised fully, close field diaphragm. - Adjust height of condenser until fuzzy edges go away from 12 sided polygon. -Open field diaphragm fully. We adjust the microscope for Kohler illumination because it ensures the best possible picture for viewing magnified specimen under the microscope.
