Exercise 13.4 A and B. Alternate Method for Preparing Lawns from Microcultures Substitute this for the Procedure on page 349 and 351. 1. Label the side edge of an agar plate with your name , equipment #, and bacteria . Each person at your table will prepare a plate to test a different 1 of these 4 bacteria: E. coli, S. marc., P. fluorescens, M. roseus . 2. sections. Then label each section with the antibiotic/disinfectant/antiseptic that will be tested. Use the order below and write small so that the writing won’t obscure the results. C lockwise, bottom up --- Antiseptic/disinfectant plate : Antibiotic plate: control (disks in the sterile H 2 O) Control (dry sterilized disks in beaker) clorox Ampicillin ethanol Penicillin Purell Neomycin Listerine Novobiocin hydrogen peroxide Tetracycline 3. Dip a kimwipe in ethanol and wipe the entire fixed-volume micropipettor barrel. This is important because the micropipettor is fat and may touch the inside of the micro-tube if care is not taken.
