E25 Problem Set 2 Solutions Please answer each question in under 50 words. If a question has a, b, c, you can use up to 50 words for each of these parts. Less is often better. 1. In general, changing the base in which position of a codon will have the smallest effect on an organism? (1 pt) Changing the base in the last position of a codon will often have the smallest effect on an organism. Most amino acids are encoded by more than one codon sequence. Inspection of the codon sequences specifying the same amino acids shows that they usually only differ in the last base. 2. What microorganism is the “workhorse” of biotechnology and why? Use the terms: doubling time, transformation . (1 pt) The microorganism that is the “workhorse” of biotechnology is E. coli. It can be easily cultured using a simple medium and grows rapidly with a doubling time of around 22 minutes. Also, it can be easily transformed with foreign DNA making it useful in recombinant DNA experiments. (S. cerevisiae or yeast is also acceptable) 3. Which mutation in the genome of an organism would you expect to be more stable, a short tandem duplication, or a large tandem duplication? Why? (0 pt) (Not grading this problem) 4. Saccharopolyspora erythraea is the bacterium that makes the antibiotic Erythromycin. Somebody isolates a mutant strain that no longer makes antibiotics. They figure out which gene is mutated (assume this gene is now totally dysfunctional and that there are no other mutations in the organism). They claim that this proves that the mutated gene has evolved specifically for making erythromycin. What is wrong with this analysis?
