Lac Operon Notes Parts of the Lac operon: LacI = codes for repressor protein (R) Promoter = P Operator = O LacZ = Z - structural gene, codes for LacY = Y - structural gene, codes for lactose permease LacA = A - structural gene, codes for a transacetylase (don’t know what this for) In the ABSENCE of lactose, the repressor is normally bound to the operator, turning OFF the Lac operon. In the PRESENCE of lactose, lactose binds to the repressor and causes an allosteric change, thereby allowing the repressor to unbind the operator and turning ON the Lac operon. Thus, the Lac operon is able to metabolize lactose in the presence of lactose, and is kept off in the absence of lactose. Terminology: R+ = normal (wt) repressor R- = mutant repressor, can’t bind to operator R S = super-repressor, can’t removed from wt operator P+ = normal (wt) promoter P- = mutant repressor, can’t transcribe genes O+ = normal (wt) operator O- = mutant operator, repressor can’t be bound Z+ = normal (wt) LacZ gene Z- = mutant LacZ gene, this can’t be transcribed Y+
