Jamie Kooser Exercise 5-2: Questions #1, 2, 3 Question 1: (a) The uninoculated controls were negative controls and the purpose that they served was for color comparison to the tubes that were used in the experiment and to ensure that the media is sterile. (b) The purpose that the PR base broth served was color change due to fermentation, which is caused by a lack of a sugar in the broth. Question 2: (a) Reversion occurred in this scenario because acid oxidizes quicker due to the small amount of glucose that is undergoing degradation. This also occurred because oxidation reactions become limited and maintain an anaerobic environment. Question 3: (a) The result of the inoculated PR broth is a false negative. (b) The false result is caused by poor sensitivity of the test system. Exercise 5-4: Questions #3, 4 Question 3: Some other consequences that might result from electron carriers in the electron transport chain being bypassed include an increase in heat and a decrease in the amount of ATP being produced due to the denaturing factor that heat provides. Question 4: A false positive from the reaction between the inoculating loop and hydrogen peroxide would be caused by poor specificity because of the sensitivity that allowed for the reaction to take place caused by the loop which give the enzyme catalase result seen in this experiment.
