Discussion: In this lab, we performed different methods of purification to purify pig heart homogenate down to GOT. We first did heat denaturation, which is good for large volume purification. This step did not purify very much as our %yield did not go lower than 100% following the heat step (from FR-I to FR-II). Also, the fold purification increased only a little bit. On day 2, we added different amount of salt, ammonium sulfate (SAS), to find out how much salt we should add on the next day for maximum recovery of GOT activity. We found that 55% SAS was the highest volume that gave us <20% GOT precipitated and above 75% SAS for >80% GOT precipitated. These values of SAS were used on day 3 to find the volume of SAS to be added. After the second salt cut with SAS we obtained AS-II, and then calculated the GOT activity of it, which was 2876.2U and 51.2% of total GOT activity. This value of % precipitated is similar to the trial from day 2. The sum of the total activities of AS-I, AS-II, and AS-III theoretically
