The examination dependsbasically on contrast and adequate lightning. One further factor involved is that, givenadequate lightning of and contrast in the structure to be examined, it must be possibleto separate them from each other. This is known as resolution(revolving power). Imageclarity depends on the microscope’s resolution which is the ability of the lens system todistinguish between two adjacent objects. If two objects are moved closer and closertogether, there comes a point when the object are so close together that the lenssystem can no longer resolve them as two separate objects. The distance betweenthem, at which they cease to be seen as separate objects, is referred to as the resolvingpower of the optical instrument.It therefore should be appreciated that only a properly stained specimen, which iscritically illuminated and examined, will give adequate resolution of the image.Furthermore, it should be realized that improperly adjusted microscopes or poorlyprepared specimens can produce artificial artefacts in the image that appear to be partof the object being examined.The microscope is an expensive piece of laboratory equipment that is extremelysensitive and yet if used intelligently, will give of years of flawless performance. It istherefore worth spending some time to understand how it works and how to use itproperly.
