Ultraviolet light causes damage to DNA by causing adjacent bases to become crosslinked. This is especially problematic for thymines, which tend to dimerize by UV treatment. b. You find that by treating the UV- damaged DNA with dNTPs, ATP and proteins isolated from nuclei of human cells restores the transformative effect of the DNA from the green microbe. Explain why, and what are the types of proteins that are responsible, and why are ATP and dNTPs necessary? This is due to the factors involved in repairing UV- damaged DNA, the nucleotide excision repair pathway. This pathway uses a protein complex (XPC- Rad23B) to recognize the distortion created by thymine dimerization. Then endonucleases (XPG and XPF) cut the 3’ and 5’ sides of the damaged DNA to remove the damaged nucleotides. Finally DNA polymerase using dNTPs, and DNA ligase using ATP, repair the damaged DNA. 2a.
