The purpose of iodine in the gram stain is to act as a mordant, it binds the dye to the cell. 2. Describe two reasons that can explain a gram positive culture appearing gram negative. Two reasons why a gram positive culture could appear negative is if you over decolorize in the gram stain procedure or if the stain is older than 18 hours, the crystal violent complex can be lost when they are old. 3. What is the color of a gram positive organism? Which dye is responsible for that color? Name one gram positive bacteria and indicate its shape. The color of the gram positive appears purple. The crystal violet dye is responsible for this color. One gram positive bacteria would be Bacillus anthracis , which is rod shaped. 4. What is the color of a gram negative organism? Which dye is responsible for that color? Name one gram negative bacteria and indicate its shape.
