Microbiology Day 4 Listen to Lecture until 10:20 Enzyme have Km for particular substrates, having different affinities -Antibiotics will have different affinities for different enzymes/targets. So when using it in his, there's a few concerns: A) Sometimes the target is specific to the bacteria, i.e. Penicillin binding proteins (PBP's), found in bacteria. This means they won't harm the host. But, antibiotics will kill natural microflora in host which is a problem. B) Common metabolic pathways, i.e. DNA synthesis, since you use many of the same enzymes, your using different Km so affinity of host enzyme should be much less, so it has a larger Km than prokaryotic enzyme, so dose is important. i.e. AZT is a nucleotide that inhibits reverse transcriptase system of HIV (RNA-dependent DNA polymerase). -This shouldn't be considered an antibiotic, but now people see it as anything that kills a disease- causing agent. -Has a very high Km for human DNA polymerase, so it will have a low effect on host. Therefore, antibiotic aren't all or none.
