Virtual Lab 1) Traditional testing often fails because it is hard to grow some pathogens on a medium that allows for easy reading and identification. Some species also cannot easily be differentiated from close relatives. In these cases it is much easier to use molecular methods. 2) You can increase the sample of bacteria by plating it on another medium and allowing it to incubate. 3) In order to extract the bacterial DNA, the first step is getting rid of the cell wall by adding proteolytic enzymes which will break apart the wall. It can take a number of hours to take place. 4) The proteolytic enzymes must be deactivated because more enzymes are going to be added. They can be denatured by heating them in a water bath at 100˚C. 5) The bacterial DNA is what is needed. It can be extracted by centrifuging the tube so that the unwanted contents settle to the bottom. The DNA will remain in the liquid. 6)
