BIO 206L: Exercise 10 Lab Report 1. Describe the structural features of pLAF (e.g, lac operon, ORI, etc.). 2. The mini-prep DNA procedure is called the “Alkaline Lysis” prep. Explain what causes the cells to lyse during the procedure. When SDS is added to the solution this dissolves the lipid membrane of the cells causing them to break open (lyse). 3. Explain what role of alkalinity (high pH) is and how this affects plasmid DNA. How does it affect chromosomal DNA? What happens when alkalinity is brought down to neutral pH? How does this affect the plasmid DNA? How does the affect chromosomal DNA? 4. (A.) Define what a restriction enzyme is and what it does.
