v.002 1 Experiment #8: Column Chromatography (Wednesday Oct. 30 th - Tuesday Nov. 5 th ) Derived in part from Gilbert, J. C., Martin, S. F., Experimental Organic Chemistry, A Miniscale and Microscale Approach, 5th ed, p. 192- 194 In this laboratory session the technique of column chromatography will be utilized in order to separate a mixture of two compounds: fluorene and 9-fluorenone. The purity of the isolated compounds will be assessed by TLC and melting point analysis . Textbook Reading G&M pg. 179-184 (Chromatography) G&M pg. 179-184 (Column Chromatography) Some Key Terms (in the context of this experiment) chromatography, TLC, column chromatography, mobile phase, eluent, stationary phase, adsorbent, polarity, equilibrium constant, partition coefficient, developing chamber, solvent reservoir, retention factor (R f ), normal-phase chromatography, reverse-phase chromatography, dry-packing, wet-packing Video Instruction You should view these videos as part of your pre- laboratory preparations. Be aware that these videos will show you a general technique but the compounds, solvents, and quantities are not the same as those you will use in the laboratory. Use these videos to become familiar set-ups and procedures, but think carefully about how your experiment may differ from the video. In your notebook, you must include a diagram of the column chromatography apparatus with labels. A flow chart is optional but it can significantly aid your ability to efficiently carry out the experiment. Important Announcements You must bring your NYU ID card to lab with you.
