Sterile Technique When working with cultures of bacteria, good technique is essential. We carry bacteria on our own hands, and a wide array of bacterial species are present in the soil and water that we encounter in our daily lives. We do not want to contaminate our laboratory cultures with these visitors. Such contamination would compromise your experimental findings. Your TA will demonstrate the basics of good sterile technique. Keep in mind that there are various levels of sterile technique and as you take other classes and/or gain experience you will likely learn much more about sterile procedures. The following is a general protocol to use when working with bacteria. Before beginning, wash your hands (and/or put on a pair of gloves) and make sure that the area in which you will be handling the bacteria is clean. Disinfect your work area with 70% ethanol from the spray bottle that has been provided and wipe it dry with a clean paper towel. When transferring sterile water or bacteria‐containing cultures, minimize the time when tube caps are off and Petri dish lids are raised.
