Using the polymerase chain reaction, we will amplify two DNA regions, including the COI gene from insects to verify we had successful DNA extraction and the 16S ribosomal DNA specific to Wolbachia to determine if the insect is infected. We will use electrophoresis to amplify the DNA of the insects. Since DNA is negatively charged, the DNA molecules will be pulled towards the positive pole. The longer molecules will go slower and stay closer to the negative pole, while the smaller molecules will go faster and be closer to the positive pole.
