Enzymes Purpose : To gain an understanding of enzymes, their activity, and how temperature, pH, enzyme concentrations affect reaction rates. Hypotheses: The 2x concentration would have the most enzymatic activity, followed by the 1x concentration, and lastly the ½ x concentration with the least amount of enzymatic activity. The 100 C sample will have the most enzymatic reaction, followed by 65 C, 55 C, 37 C, 22 C, and lastly 0 C with the least amount of enzymatic activity. pH 7 will have the most enzymatic activity, followed by 8.5 and 5.5 (with equal amounts of enzymatic activity), and 4 with the least enzymatic activity. Method Outline: 1. Label two large test tubes with #1 and #2 directly on the tubes. Tube #1 will contain the substrate, and Tube #2 will contain the enzyme (peroxidase) 2. Covering tubes with a piece of parafilm, vortex tube #1 and tube #2 separately. Pour all of tube #1 into tube #2. Cover the tube with a piece of parafilm and vortex for about 5
