Unformatted text preview: 2/11/08 Lab 3: Fluorescence Microscopy PURPOSE: The purpose of this lab is to familiarize the students with fluorescence microscopy, and various staining tactics used in combination with fluorescent materials to get a better look at organisms otherwise difficult to see. ABSTRACT: The students were split into two groups and assigned different procedures to work on at the beginning of this lab. The first group was in charge of double-staining nuclear material. 400 L of rhodamine phalloidin solution was added to the given cells, and placed in a covered location for 30 minutes. After this DAPI solution was added for 5 minutes, and then it was washed 3 times with PBS. The cover slip was then mounted on a slide and viewed with rhodamine and then UV filters. The second group was tasked with double-staining the nucleus and microtubules in their cells. 400 L of mouse anti-tubulin was added and allowed to sit in a covered area for 30 minutes. cells....
