3/17/08 Lab 7: Plant Development Lab PURPOSE: The purpose of this lab is to observe several Arabidopsis thaliana variations and mutations to determine the expression of traits such as leaf size, shape, pigment, and trichomes. ABSTRACT: This lab was broken up into three parts, all dealing with observation of the traits of Arabidopsis thaliana . In the first part, 0.5 mL of X-Gluc stain solution was added along with a few seedlings into a 1.5 mL microfuge tube. This tube was placed in a 37 °C incubator and allowed to sit for about 2 hours. After the time was up, the seedlings were separated from the stain with a micropipetter, and 1 mL of 25% ethanol was added and allowed to sit for 5 minutes. After this, 50% ethanol was added, then 75% and 95%. Once all of this was done, the seedlings were observed under a microscope. The second part of this lab had students observe and sketch the various mutant seedlings provided by the instructor. The third and final portion of this lab had students remove 5 seedling leaves and place them into a microfuge
