Cell Biology 03-240 Assignment 1 1. Let us assume that know how to measure the levels of different lipids PC, PS, PE, and PI levels in cultured cells. a) (10 pts) You add a phospholipase, a water-soluble protein enzyme that cleaves off the phosphate head groups of phospholipids, to the culture medium. You measure the levels of these four phospholipids and compare it to cells where you have not added the enzyme. Do you think the levels of all four phospholipids will be reduced to an equal extent compared to their initial levels? If yes, briefly explain why. If not, which ones will be reduced more compared to others? Briefly explain why. Phosphatidyl choline (PC) will be reduced more compared to others. Because it is a water- soluble enzyme, it cannot cross the membrane. So it will preferentially cleave of phospholipids on the exoplasmic surface. Since PC predominates on the exoplasmic surface, more of those will be cleaved.
