Essay 9 Describe the structure of an enzyme at the molecular level and precisely how it mediates a chemical reaction. Please include a description of free energy and energy of activation as they relate to enzyme mediated reactions. Precisely how do enzymes reduce the energy of activation. Protein Structure : Primary - amino acid sequence - (peptide 50 amino acids or less) all subsequent structure is a consequence of primary structure. Secondary - alpha helix or beta pleated sheet results from backbone N/O/H hydrogen bonding between C=O and N-H Tertiary Structure - Hydrophobic with self and forced internal to water Hydrophilic with self (+ to -) and interacts with water ionic bonds, hydrogen bonds Disulfide - covalent internal linkage Quaternary – two or more proteins attached to each other, generally by disulfide bond Central Concept in Biology : enzymes are key element in production of toads from genes because they control all reactions, including synthesis of all other molecules. They control synthesis by reducing the energy of activation . Reactants fit into close association with the three dimensional shape of the protein.
