The ATG sequence that is the start codon of the coding sequence is marked on the figure by an arrow just inside the left hand BamHI site. Digest 1. BamHI 2. AatII Fragment Size 2000 bp, 3500 bp 1000 bp, 4500 bp The first digest with BamHI will act as a control just to show that the 2000 bp cDNA fragment is present. The second digest with the AatII would be enough to show the orientation of the cDNA
