8)Predict what effects, you would expect the following to have on the migration of LDH in the gel (compared to the gel you ran), and briefly explain why:
a)
By increasing the agarose percentage to 1,2% , it will slow down the speed at
which the molecules travel through the gel , since the pore of the agarose
become smaller due to the increased percentage. Some large molecules will
not be able to migrate since
the pores will become too small. (1)
b)
By decreasing the pH. to 6 , it will affect the migration since it will affect the
overall charge of the protein.,
some amino acids will get protonated thus
becoming positively charged, For example the
amino histidine will have a
positive charge at a pH. of 6. This will have a consequence in the migration
pattern since it will decrease its migration distance since the protein is more
positively charged.
c)
A decrease in voltage will slow the migration speed of the protein since the
anode on the other side of the gel won’t pull
the negatively charged particles
as strong as with a higher voltage
