Answer: Mutant a- When sugarose was added to the media, there was no SugB activity but SugA activity was intact. You create partial diploids using an F’ factor carrying the Sug operon. Here are your results. See Table 2 in the course-pack for this problem. Hint: for parts (c)-(f), rather than jumping in and trying to answer part (c), try to figure out what each mutation is. From the first table you can figure out whether the mutation causes uninducible or constitutive expression. Mutation a = SugB structural gene Mutation b = SugA structural gene Mutation c = constitutive activity – either “broken” operator or repressor Mutation d = uninducible activity – probably “broken” promoter, super-repressor, or activator Mutation e = constitutive activity – either “broken” operator or repressor (c) Which mutation is most likely to inactivate the promoter for the Sug operon? Answer: Mutant d- A mutation that causes inactivation of the promoter prevents the structural genes from functioning, thus the operon becomes uninducible. Mutant d- is the only one that does this.
