Lab 7 Page 4 Lab 7 Exercises Artificial Epidemic We’ll see who contracts Jawa Fever during the Artificial Epidemic via Hand Shaking. This is a Propagated Epidemic, which is characteristic of a contagious Microbial Disease. Typical Propagated Epidemics would be Influenza or Polio. You contract these from Direct Contact with another Person or -- more to the Point -- from Direct Contact with Bodily Fluids from another Person. Each Student will choose a Tube of Bacteria. There are 23 Tubes of E. coli and one Tube of Pseudomonas putida (which produces fluorescent Siderophores on F Agar). These 24 Tubes of Bacteria look identical. The Materials (per Student): • One F Agar Plate (marked into Quadrants) • One Tube of Bacteria • One Latex Glove • One Swab The Set-Up 1. Label your Nutrient Agar Plate with your Name. This is Important. 2. Divide the Bottom of your Nutrient Agar Plate into Quadrants and label these Quadrants I, II, III, and IIII. Viewed from the Bottom, the “I” goes on the Upper Right Quadrant, “II” on the Upper Left, “III” on the Lower Left, and “IIII” on the Lower Right. 3. Obtain a Tube of Bacteria. You won ʼ t know if you ʼ ve got E. coli or Pseudomonas putida until next Lab. 4. Obtain a Small, Medium or Large Latex Glove. Put the Glove on your Right Hand (even if you ʼ re Left Handed) 5. Obtain a Sterile Swab. You can toss the Swab Packet into the Trash at the End of Lab. The Swab itself will go into the Stainless Steel Pitcher on the Waste Cart. 6. Unwrap the Swab, stick it into your Tube of Bacteria and use it to stir-up your Bacteria (as they say in Jamaica, “Stir it up!”) This will be the Signal that you are ready to Start Please do not shake Hands until told to do so by your Lab Instructor.

Lab 7 Page 5 The Epidemic (“Utini!”) 1. At the Signal from your Lab Instructor, take your Swab and slather your Bacterium all over your Glove. Save this Swab by leaving it in your Tube. • Rub your Fingertips across your Palm. • Use your Index Finger to make a Quarter-Sized Smear on Quadrant I. Your Glove will absorb an astonishing amount of the Bacterial Culture, so you ʼ ll have to repeatedly swab your Glove to get it good and goopy. You ʼ re also going to discover that these Cultures stink to High Heaven. At this Point, only 1 Student should have Jawa Fever. 2. At the Signal from your Lab Instructor, re-slather your Glove. • At the Signal from your Lab Instructor, shake Hands with your Lab Partner. This won ʼ t work if you formally shake Hands. You ʼ ve got to get Rude, Crude, and downright Lewd to effectively transfer Bacteria from Glove to Glove. But no High Fives, Please. (Splatter) • Now, back to your Plates, rub your Fingertips across your Palm, and then take your Index Finger and make a Quarter-Sized Smear on Quadrant II.
