. 5 Causes of Hemolysis:(cont.) 1- In vivo-hemolysis In-vivo hemolysis is caused by : a) antibodies, b) biochemically through medications, by toxic substances, through hereditary factors (e.g. haemoglobinopathies), through enzyme defects (choleric jaundice) c) infections (e.g . malaria). When suspecting in vivo hemolysis the plasma should be checked to exclude the possibility of additional in vitro hemolysis caused by the coagulation process.

2-In vitro-hemolysis Several factors causing in vitro hemolysis are encountered during the venipuncture procedure. 1. Residual alcohol at the skin puncture site. 2. “Milking” of the puncture site. 3. Shaking the blood too vigorously. 4. Drawing blood through a hematoma or from a vein with a hematoma. 5. Using a large volume tube with a small diameter needle or in a weak vein. 6. Improper centrifugation time and speed. 7. Positive or negative pressure in sample tube. 8. Blood dilution with hypotonic solution.
