If you ʼ re a Clan of Four, proceed to Page 7. If you have a Clan of Three, Spread Wallah replaces both N-Spreader and MacSpreader. Spread Wallah will first do all the Nutrient Agar Plates with one sterile Spreader, then do all the MacConkey Agar Plates with a new sterile Spreader. Advance to Page 8.

Lab 6 Page 7 Clan of Four Spread Plating 1. Vortexer positions the Dilution Series, Turntable, Nutrient Agar Plates, MacConkey Agar Plates, Pipetter , N-Spreader and MacSpreader for maximum Ergonometric Efficiency. 2. N-Spreader and MacSpreader each remove a sterile Spreader from the Packet. You ʼ ll each use one (1) sterile Spreader for the entire Experiment (unless you screw-up). Remove the Spreaders from the Packet very carefully. V 3. Vortexer vortexes the 10 -7 Dilution Tube and positions this Tube so P Pipetter can transfer 100 μ l (P200 at 1 | 0 | 0) onto the Surface of N-Spreader’s “8” Nutrient Agar Plate. N A. N-Spreader places this “8” Nutrient Agar Plate in the Center of the Turntable, Bottom Side Down/Lid Side Up. B. N-Spreader lifts the Lid, slowly spins the Turntable and uses their Spreader to evenly spread this Sample across the Surface of their “8” Nutrient Agar Plate. Spread this Sample thoroughly, but expose the Surface of this Nutrient Agar Plate to the Air for as short a Time as possible. C. N-Spreader replaces the Lid and moves this Plate somewhere else. V 4. Meanwhile, Vortexer again vortexes the 10 -7 Dilution Tube and positions this P Tube so Pipetter can transfer 100 μ l (P200 at 1 | 0 | 0) onto the Surface of MacSpreader’s “8” MacConkey Agar Plate. Mac A. MacSpreader places this “8” MacConkey Agar Plate in the Center of the Turntable Bottom Side Down/Lid Side Up. B. MacSpreader lifts the Lid, slowly spins the Turntable and uses their Spreader to evenly spread this Sample across the Surface of their “8” MacConkey Agar Plate. Spread this Sample thoroughly, but expose the Surface of this MacConkey Agar Plate to the Air for as short a Time as possible. C. MacSpreader replaces the Lid and moves this Plate somewhere else. 5. Repeat Steps 3 and 4 (Above) for each successive Pair of Nutrient Agar and MacConkey Agar Plates. 6. Place your Spreader in the “Microtiter Pippete Tips” Waste Beaker. 7. Tape your Nutrient Agar Plates together in two Stacks of 3 Plates each (or one Stack of 6) and incubate them at 30°C for 48 Hours . 8. Tape your MacConkey Agar Plates together in two Stacks of 3 Plates each (or one Stack of 6) and incubate them at 37°C for 24 Hours .
