Once the reagent is applied, one must continue to shake at the same temperature for about 2 minutes. After the preparation of the gel tray, and once the flask is cool, pour the mixture inside the apparatus, and ensuring no bubbles form in the agarose. It is necessary to wait for around 20 minutes allowing the gel to solidify before adjusting it to the proper orientation in the box. The gel must be submerged by 1x TAE/TBE buffer before gently removing the comb. Then the gel will be ready to undergo electrophoresis.

We prepared samples in 200ul tubes, labeling and adding the reagents described in the table on page 63 in the lab manual. Once these samples are loaded, the lid should be closed, power supply connected, and switched on at 110 volts. Then electrophorese for one hour until the dye has traveled 2/3 the length of the gel. The product can now be turned in to the lab instructor who will have it photographed for us. The lab work area is cleaned up before leaving the lab. Good handwashing should be practice at all time because of bacterial infection and proper hygiene. Results and Discussion
