crossed-over to The Dark Side. When grown on EMB Medium E. coli produces Purple Colonies and a characteristic Fly-Butt Green Metallic Sheen on the Surface of the Medium. E. coli is one of the most intensively studied of all Species. It has been the Lab Rat for Classical Experiments in Microbiology and Molecular Biology. All Escherichia in Nature are entirely Symbiotic, occurring in large Numbers in the Gut of Warm-Blooded Animals. Escherichia is not normally Pathogenic, but can cause serious Infections when it gains Access to the Bladder. E. coli can also cause Peritonitis if the Intestine is punctured -- say by a Gun or Knife Wound -- and the Intestinal Contents are released into the Peritoneal Cavity. You should treat Escherichia as a Pathogen. E. coli is on EMB Medium. The Production of buttloads of Acid results in the Precipitation of the metachromatic Dye Methylene Blue, making the Colonies appear dark Purple and the Agar to take-on a Fly-Butt Green Metallic Sheen. When grown on Nutrient Agar, E. coli Colonies are Clear-to-White and indistinguishable from Enterobacter aerogenes Colonies. The IMViC Test is also used to distinguish E. coli from Enterobacter . The IMViC Test consists of the combined Results of four Biochemical Tests: Indole, Methyl Red, Voges-Proskauer and Citrate (the ”i” was inserted to supply a Vowel). Aseptic Technique is the sine qua non (Latin for “without this nothing”) of Microbiology. The Techniques you’ll start learning Today are absolutely Essential for all of our Exercises and for identifying your Unknown Cultures. The basic Idea is to use Sterile or sterilized Instruments and to minimize the Time a Serological Pipette, Test Tube or Petri Plate is uncovered. We want to move our Bacteria from Container to Container without letting any other Bacteria into these Containers and without letting our Bacteria out of these Containers. The Terms “Aseptic Technique” and “Sterile Technique” are informally used interchangeably by most Microbiologists and Cell Biologists. However, the two Terms have subtly different Meanings: Aseptic Technique involves working with only the Organisms of Interest. We ʼ ll be using Aseptic Technique to transfer E. coli from one Petri Plate to another later on in Lab. Sterile Technique
