Again, 10 Tubes can be used. • Each of 3 Standard Test Tubes of Lactose Broth is inoculated with 0.1 ml of this Water Sample. Yes, you can use 10 Tubes here, too. • These 9 Tubes are then incubated at 35° for 24 Hours ± 2 Hours. - Turbidity (L= mixed-up) (“Cloudiness”) and Gas Production (a Bubble of Hydrogen in the Durham Tube) in any Tube is Positive for the Presence of Coliforms. Each Set of Tubes generates a Number between 0 and 3 so an MPNTest will generate three Numbers and these can be used to determine MPN from Statistical Charts.

You might notice that the First Stage of this Test (The Presumptive Test) is simply an MPN -- but all Tubes must test Negative. • Presumptive Test - Tubes containing Lactose Broth are inoculated with 10 ml, 1.0 ml or 0.1 ml of Sample Water - Incubated at 35° for 24 ± 2 Hours - Gas Production is Presumptive of the Presence of Coliforms • Confirmed Test - The most Dilute Gas-Producing Tubes are streaked onto Eosin-Methylene Blue (EMB) Plates - Incubated at 35° for 24 ± 2 Hours - Dark-Centered Colonies (frequently surrounded by a Fly Butt Metallic Green Sheen) confirms the Presence of Coliforms If you ʼ re in MIC 102L you know these EMB Test Results indicate the Presence of E. coli . • Completed Test - Tubes containing Lactose Broth are inoculated with Dark-Centered Colonies • Incubated at 35° for 24 ± 2 Hours • Gas Production and Acid Production is Positive
