2. Turn your Petri Plates Upside-Down and mark a “T” on the Bottom of all four (4) of your Petri Plates. The Area “above” the Horizontal Line of the “T” is called the “A” Section. The Area to the Right of the Vertical Line of the “T” is called the “B” Section and the Area to the Left of the Vertical Line of the “T” is called the “C” Section. _____________________________________________________________________ I've just about had enough of you. Lab 1 Page 7 3. Label the Bottom of your Plate(s) with your Name, “Skin” (YGC) or “Rectal” (Nutrient Agar and EMB) and the Date. Label around the Edge of the Bottom (or better yet on the Sides of the Bottom) so your Writing doesn ʼ t interfere with your View from the Bottom of any Colonies that grow on the Plate. 4. Keep these Plates Lid-Side Down. 5. Sterilize your Loop in your Bunsen Burner. 6. Carefully lift the Bottom of the Mother Plate. 7. Cool your Loop by touching the Surface of the Agar in the Bottom of this Mother Plate. It is extremely Important that your Loop be Cooled before you proceed to the next Step. You should hear a satisfying “Steak House Sizzle” as the Hot Loop hits the Agar (which is mostly Water). Use a Portion of the Mother Plate (probably the “C Segment” (3rd Streaking Segment) with as few Colonies as possible 8. Touch your Loop to the Edge of an Isolated Colony on this Mother Plate. You needn ʼ t scoop. Even though it doesn ʼ t look like it, merely touching the Edge of a Colony will give you beacoup Bacteria. Pretend you are using your Loop to sample some really primo, ichiban Wasabi that your Roommate has brought back from Kyoto and you don ʼ t want to have your Head blown off. 9. Place the Bottom of the Mother Plate back on its Lid. 10. Lift the Bottom of one of your Petri Plates. Streak your Loop back and forth over the Agar above the Top of the “T”, or roughly the upper 1/3 of the Plate (which we’ll call the “A” Segment). Remember not to lean directly over the Plate. Your Hair is a Microbial Rainforest. Glide along the Surface and try not to break through. Try to make your Streaks as close together as you can. The more Streaks you can make, the better the Separation of Bacteria. It ʼ s okay if they overlap somewhat in this ”A” Section. 11. Re-sterilize your Loop in the Bunsen Burner. 12. Cool your Loop by touching the Edge of the Agar in an unused Part of your Plate. The Intersection of the Lines you drew to make the “T” works really well for cooling your Loop. It ʼ s easy to see and you can use it multiple Times.

Lab 1 Page 8 13. Rotate your Plate by 90 Degrees (1/4 Turn) such that you can glide your Loop through the “A” Section at the Top of the “T” once and then streak in a Zig-Zag across the Side of the Base of the “T”, or roughly another 1/3 of the Plate (which we call the “B” Section).
