3.  Predict the most likely effect of the following mutations on a typical cell. Explain your answer.   (The transport into mitochondria and peroxisomes will be covered on the  next week’s lectures).   (A) Replacement of the second amino acid in Urate Oxidase (a peroxisomal protein)  with an Arg instead of a Lys.  1. This replacement will result in a missense mutation in the peroxisomal  protein. Urate Oxidase normally acts as an oxidative enzyme when it is  localized by a specific signal sequence, but since it is a missense on the  second amino acid with an amino acid that has the same charge as the  original it can still function but improperly. Some possibilities include,  oxygen being built up in the peroxisome or inability of proteins to be  imported into the peroxisome because the specific sequence of (Ser-Lys- Leu) changed to (Ser-Arg-Leu). This sequence has an interruption and  prevents the protein from being recognized by soluble receptor proteins.  (B) Deletion of the first hydrophobic domain of a subunit of the mitochondrial DNA  encoding the Complex I electron carrier NADH Dehydrogenase.
