Lab 1 Page 10 Isolations Isolation of Pseudomonas (Enrichment) Materials [per Student] • Sterile 16mm Test Tube • Sodium Benzoate Broth • Soil Sample Procedure 1. Swirl the Sodium Benzoate Broth and add ~5 ml (~ 3 Fingers) of this Sodium Benzoate Broth to a labeled 16mm Test Tube. 2. Add a very small Pinch of Soil to the Sodium Benzoate Broth in your Test Tube. (A “pinch” is less than 0.5 g.) 3. Incubate your Test Tube at 25 ° C until the next Lab. Isolation of Bacillus (Direct Isolation and Selection) Materials [per Student] • Sterile 16mm Test Tube • Test Tube Rack • Distilled Water • Soil Sample • 80°C Water Bath (the steaming one over by the Windows) • Nutrient Agar Plate Procedure 1. Add ~ 5 ml (Three Fingers) of non-sterile, De-Ionized Water to a (labeled!) Sterile Test Tube. 2. Add a Pinch of Soil to the Test Tube and Finger Vortex the Soil Suspension 3. Pasteurize the Soil Suspension in an 80 ° C Water Bath for 20 Minutes. Remember: 80°C is really Hot! 4. Allow the Soil Suspension to cool-down enough to handle it. 5. Finger Vortex the Soil Suspension so that the Soil is fully suspended. 6, Use your Sterilized Loop to streak a Loopful of your Pasteurized Soil Suspension onto a (labeled!) Nutrient Agar Plate. 7. Incubate your Clan’s 4 Plates at 30 ° C for 48 Hours.

Lab 1 Page 11 Isolation of Skin Bacteria (Direct Isolation) Materials • “Patient” Skin Swab Plate (1 Plate per Clan) • YGC Plate (1 Cream-Colored Agar, Red Stripe Plate per Student) Your Patient Skin Swab Plate is a Medium that doesn ʼ t look like YGC and that doesn ʼ t distinguish Micrococcus luteus from Staphylococcus epidermidis like YGC. So the Identify of your Patient Skin Isolate will only be revealed after you ʼ ve incubated the YGC Plates your Clan streaks Today. Procedure 1. Each Clan gets one (1) “Patient” Skin Swab Sample Plate. 2. Streak a YGC Plate from an isolated Colony on your Patient Plate. 3. Incubate your Clan’s 4 YGC Plates at 37 ° C for 48 Hours. 4. Save your Patient Skin Swab Plate at 04°C. Isolation of Enteric Bacteria (Direct Isolation) Materials • “Patient” Rectal Swab Plate (1 Plate per Clan) • EMB Plate (1 Burgundy-Colored Agar Plate per Person) • Nutrient Agar Plate (1 Nutrient Agar Plate per Person) Your Patient Rectal Swab Plate is a Nutrient Agar Plate, which doesn ʼ t distinguish Enterobacter areogenes from E. coli like EMB. So the Identity of your Patient Rectal Isolate will only be revealed after you ʼ ve incubated the EMB Plates your Clan streaks Today.
