For biochemistry lab we did gel filtration chromatography purification of LDH. The directions I've been given say

to determine Ve the elution volume for LDH from the peak of a plot of A340/min vs fraction number. It says if ours ''didn't work out'' to consider the middle fraction of my pool as the peak. My pooled fractions were 43 through 49. I understand that the elution volume is the volume from the time the LDH was applied to when it elutes. I applied the LDH at fraction 6 and each fraction was 1 mL. Would the Ve be the highest peak at fraction 44 or would it be at the second peak above fraction 47?
