INTRODUCTION High performance liquid chromatography (HPLC) is one of the major methods of analysis of organic materials. In common with other chromatographic techniques, HPLC offers a high degree of analytical selectivity and its applications scope is not limited by sample volatility or thermal stability characteristics as is the case with gas chromatography. Liquid chromatography has been in use for many years, but it is only recently that improvements in column technology and instrumentation have been made which have enabled the technique to be adopted for high-speed analytical purposes. In its present state of development, HPLC offers a variety of highly efficient separation modes, which, in total, provide a very wide application potential. PRINCIPLES OF HIGH PERFORMANCE LIQUID CHROMATOGRAPHY The technique of HPLC involves the separation of the various constituents of a sample followed by their individual detection and measurement. The separation is achieved by a competitive distribution of the sample between two phases, one a mobile liquid and the other a stationary liquid or solid. In modern liquid chromatography the stationary phase is typically supported in a column about 25 cm long and 4 mm internal diameter. Shorter columns (150 mm to 33 mm) are now also routinely used, as with wider and narrower columns (7 mm to 1.2 mm). High separating efficiency has been achieved by the optimisation of column parameters and, in particular, of the particle size of the column packing. In general, it has been shown

Experiment 5 High Performance Liquid Chromatography 2 that a reduction in particle size of the column packing results not only in an improvement in column performance but also in an increase in the pressure required to obtain a satisfactory flow of mobile phase through the column. For this reason, modern liquid chromatography is often referred to as `high pressure liquid chromatography' but, in fact, provided that the column packing is of uniform particle size, only moderately high pressures are required to give the necessary flow rate of 1-2 mL min -1 . (As a guide, typically a 25 cm × 4 mm column packed with l0 µ m particles requires about a pressure of 30 atm for a flow of 1 mL min -1 of hexane and about 50 atm for a similar flow of a more viscous solvent such as water). Improvements in column technology have meant that these days a 33 mm × 7 mm I.D. column with 3- µ m packing is commonplace and is more than capable of out performing a 25 cm × 4 mm I.D. column. The basic apparatus required to perform HPLC is shown in Figure 1. Figure 1. Schematic arrangement of HPLC components. The mobile phase is forced through the separating column by the pump. It then flows through the detector. As each sample component passes through the detector a change in electrical output is produced which is recorded on a moving chart to give a chromatogram as shown in Figure 2. Figure 2. An HPLC chromatogram. The features of the chromatogram are: 1. The time taken for a substance to pass through the column under fixed conditions is constant and is called the retention time. Comparison of retention times with those of standards enables liquid chromatography to be used for qualitative analysis.
